报告题目:Profiling Carbohydrate Utilization in Gut Microbiomes of Diverse Human Populations

The human gut, especially the large intestine (or colon), is a carbohydrate rich environment with a very high diversity of carbohydrates (also known as fibers or glycans). After ~30 years of human microbiome research, it is now widely accepted that, consuming a high-fiber diet on a daily basis is extremely critical to maintain our gut microbiota diversity and prevent various metabolic diseases. The western diets are known to be unhealthy as it lacks sufficient dietary fibers, leading to various metabolic diseases (e.g., cardiovascular disease, intestinal bowel disease, colon cancer, and obesity). This is because consuming fiber-deprived diets for a prolonged time will force gut bacteria to more aggressively digest host-derived much glycans. Mucins are secreted by gut epithelial tissues forming a physical barrier to protect host tissues from potential threats, including microbial pathogens. To fully digest diverse complex dietary fibers and host-derived glycans, hundreds of human gut bacteria dedicate up to 10% of their genes encoding carbohydrate active enzymes (CAZymes). Personalized dietary fiber intervention, by giving different dietary fibers as prebiotics to different individuals, is an effective strategy to nurture a healthy gut microbiome and thus enable personalized nutrition for disease prevention. In this seminar, I will introduce our recent bioinformatics tool development for our dbCAN software system that can profile CAZymes and glycan in microbiome samples of diverse human populations. I will also talk about the discovery of a new gut bacterial species named Limousia that uses a previously unrecognized molecular mechanism for highly organized mucin glycan degradation. After the continuous development since 2012, our dbCAN software system has become the most popular bioinformatics software tool for automated CAZyme annotation in microbiomes with a large user base.

尹燕斌教授任职于美国内布拉斯加大学(University of Nebraska Lincoln)食品科学和工程系,是内布拉斯加州食品和人类健康中心PI,研究方向为生物信息工具研发、计算和进化基因组学。2005年在北京大学生命科学学院生物信息中心获生物信息学博士学位。随后在美国纽约州立布法罗大学计算机系、佐治亚大学生物信息研究所先后任博士后和研究科学家。尹教授长期以来研究植物细胞壁和碳水化合物酶(CAZyme),开发并利用生物信息工具来自动挖掘微生物以及植物基因组中的CAZyme,促进其在生物能源,肠道菌群,和人类健康中的基础和应用研究。课题组开发的dbCAN系列工具成为最常用的CAZyme自动功能注释工具,dbCAN的三篇web server文章均为高被引和热点文章 (cited >5,000 times)。尹教授在Nature Genetics、PNAS、Gut Microbes、Nucleic Acids Research、mBio、Bioinformatics等国际重要学术刊物发表第一或通讯作者论文七十余篇,总计SCI论文120多篇,谷歌学术H-index为49,总引用超过1.2万次。2017年获得美国NSF CAREER Award奖,目前实验室的研究获得美国国立卫生研究院(NIH)R01和农业部的资助。
